電針聯合兩色金雞菊提取物對糖尿病腎病大鼠腎臟Vimentin、α-SMA、TGF-β1及p-smad2表達的影響

摘要:目的? 觀察電針聯合兩色金雞菊提取物對糖尿病大鼠腎臟波形蛋白(Vimentin)、α-平滑肌肌動蛋白(α-SMA)、轉化生長因子-β1(TGF-β1)及p-smad2表達的影響,探討其作用機制。方法? 將36只SD大鼠隨機分為正常組、模型組、電針組、兩色金雞菊組、電針+兩色金雞菊組和二甲雙胍組,每組6只。采用高糖高脂喂養聯合腹腔注射鏈脲佐菌素建立2型糖尿病模型。正常組、模型組不進行干預;電針組針刺大鼠雙側“腎俞”“脾俞”,連接電針,留針30 min;兩色金雞菊組予兩色金雞菊乙酸乙酯提取物(300 mg/kg)灌胃;電針+兩色金雞菊組予電針聯合兩色金雞菊乙酸乙酯提取物灌胃;二甲雙胍組予二甲雙胍(200 mg/kg)藥液灌胃。每日1次,治療4周。HE染色觀察腎組織形態學及纖維化程度,Western blot檢測腎組織Vimentin、α-SMA、TGF-β1及p-smad2蛋白表達。結果? HE染色結果顯示,電針組、兩色金雞菊組、電針+兩色金雞菊組和二甲雙胍組均可緩解模型大鼠腎小管擴張,減輕腎小球皺縮現象,改善炎性細胞浸潤,減少基底膜增生和融合,且以電針+兩色金雞菊組改善更明顯;Western blot檢測結果顯示,電針和兩色金雞菊提取物均可降低模型大鼠腎組織Vimentin、α-SMA、TGF-β1及p-smad2蛋白表達,且以兩者聯合效果最佳。結論? 電針聯合兩色金雞菊提取物可明顯改善糖尿病大鼠腎臟纖維化,其作用機制可能與抑制TGF-β1/Smad2信號通路有關。

Abstract: Objective To observe the effects of electroacupuncture combined with coreopsis tinctoria extract for expressions of Vimentin, α-SMA, TGF-β1 and p-smad2 in renal tissue of diabetic rats; To discuss its mechanism of action. Methods Totally thirty-six SD rats were randomly divided into normal group, model group, electroacupuncture group, coreopsis tinctoria group, electroacupuncture combined with coreopsis tinctoria group and metformin group, with 6 rats in each group. Type 2 DM model was established by high-fat and high-glucose diet and STZ injection intraperitoneally. Normal group and model group were given no treatment; Electroacupuncture was at the acupoints of “Pishu” and “Shenshu” once daily in electroacupuncture group for 30 min; coreopsis tinctoria group was given intragastric administration of 300 mg/kg coreopsis tinctoria extract for gavage; electroacupuncture combined with coreopsis tinctoria group was given electroacupuncture and intragastric administration of coreopsis tinctoria extract for gavage; metformin group was given intragastric administration of 200 mg/kg metformin for gavage. Histological changes and fibrosis degree of kidneys were observed by HE staining and expressions of Vimentin, α-SMA, TGF-β1 and p-smad2 were detected by Western blot. 糖尿病腎?。╠iabetic nephropathy,DN)已成為終末期腎病的第2位致病因素[1]。終末期腎病一旦形成,治療極為棘手。因此,及時治療DN,防止其向終末期腎病轉化意義重大。腎臟纖維化是DN主要的病理特征之一,前期研究發現,兩色金雞菊提取物對高糖高脂誘導的大鼠腎小球系膜纖維化有明顯抑制作用[2-3],而電針也可改善DN腎功能和超微結構[4]。本實驗擬將電針聯合兩色金雞菊提取物干預DN大鼠,觀察其對DN大鼠腎臟波形蛋白(Vimentin)、α-平滑肌肌動蛋白(α-SMA)、轉化生長因子-β1(TGF-β1)及p-smad2表達的影響,明確兩者是否存在協同作用,為臨床治療DN提供依據。
